A model of cholesterol 7a-hydroxylase gene (CYP7A) promoter. Two bile acid response elements (BARE) are located in the proximal promoter. Nuclear hormone receptors HNF-4 and COUP-TFII bind to BARE-II and BARE-I, respectively, and synergistically activate gene transcription. RXT/RAR may compete with HNF-4 for binding to BARE-II. The TC-rich region located between BARE-I and BARE-II may bend the DNA and loop BARE-II over BARE-I, and facilitate the interaction with TATA box binding protein TFIID and enhance RNA polymerase II activity. HNF-3 also transactivates the gene. (See Crestani el., p. 2192).
