PUFA and cholesterol repress the maturation of SREBP via the SREBP cleavage-activating protein (SCAP) that would otherwise translocate from the ER into the nucleus to activate the transcription of SCD genes by binding to SRE. PUFAs from the diet or membrane phospholipid (mPL) independently bind the putative binding protein (PUFA-BP) and the PUFA and PUFA-BP complex represses transcription of the SCD genes by binding to PUFA response element (PUFA-RE). Crosstalk between the two pathways is designated by the dashed arrow. (See Ntambi, p.1549.)
