COVER: Fluorescence photomicrographs of unesterified cholesterol in THP-1 macrophages. Cells were loaded with cholesterol using either mildly oxidized LDL (top left) or acetylated LDL (bottom left). Treatment of oxidized LDL-loaded cells (top right) with 15 mM 2-hydroxypropyl-
-cyclodextrin removed primarily the plasma membrane cholesterol and had little effect on intracellular pools. In contrast, treatment of acetylated LDL-loaded cells with cyclodextrin (bottom right) substantially removed both membrane and intracellular free cholesterol. This indicates that, unlike cholesterol from acetylated LDL, the large intracellular stores of lysosomal cholesterol from oxidized LDL are trapped within the cell.
