COVER: Expression of Rab7 and Rab9 in NPC1 cells. Human NPC1 fibroblasts expressing Rab9 (A, right) are completely devoid of filipin-positive material (A, left). Mouse NPC1 cells coexpressing human Rab9 and green fluorescence protein also exhibit a correction of their filipin-positive phenotype (B). Expression of Rab7 (C), Rab9 S21N (D), or Rab7 T22N (E) in NPC1 cells did not result in correction of the NPC1 filipin phenotype. Notably, most cells expressing Rab7 T22N were in the process of undergoing apoptosis. (See Walter, Davies, and Ioannou p. 243.)
