COVER: BC-theta is a biotinylated derivative of a cholesterol-binding protein perfringolysin O (theta-toxin). In unfixed cells, it detects cholesterol at the cell surface (top row); in 1% paraformaldehyde fixed cells, it detects cholesterol at and near the cell surface (middle row); in 4% paraformaldehyde fixed cells, it detects cholesterol-rich domains in the cell interior (bottom row). Parental (left panels) and Niemann-Pick type C (right panels) Chinese hamster ovary cells were used. (See Sugii et al.,p.1033.)
