COVER: Plasma membrane lipid composition influences amyloid precursor protein (APP) metabolism. Cholesterol-poor regions of the plasma membrane favor the proteolytic pathway mediated by the "a disintegrin and metalloprotease" (ADAM) class of proteases, which cleave APP at the site and generate the neurotrophic APPs fragment. In contrast, cholesterol- and sphingolipid-rich regions of the plasma membrane, or lipid rafts, favor the proteolytic processing of APP at the site by the -secretase/presinilin-1 (PS-1) protease complex and at the site by the beta-site APP-cleaving enzyme (BACE), to generate the neurotoxic amyloid- peptide (A) shown as a solid rectangle.
