COVER: Schematic superposition of the -helical isoprenyl-binding pocket of farnesyl transferase (orange), geranylgeranyl transferase-1 (green), and Rab geranylgeranyl transferase (yellow). A bulky residue (tryptophan) in farnesyl transferase shortens the pocket and sterically hinders binding of geranylgeranyl pyrophosphate (GGpp; red) but not farnesyl pyrophosphate (Fpp; blue). At the equivalent position, both geranylgeranyl transferases contain a smaller residue, which lengthens the pocket and allows binding of geranylgeranyl pyrophosphate. (See Leung et al., p. 467.)
