COVER: Lipotoxicity may be induced by death receptor signaling via alterations in expression and/or activation of these proapoptotic mediators. Ligation of the death receptors, Fas or TRAIL-R1/R2 by their ligands, induces the formation of an intracellular signaling platform termed the death-inducing signaling complex (DISC), which via generation of active caspase 8 results in direct or indirect activation of effector caspases 3, 6, and 7, which execute the final steps of cell death.
